چکیده

Chromosomal aberrations are of great concern in dairy animals worldwide as they are usually associated with reduced fertility or infertility. There are multiple reasons which may cause chromosomal aberrations during mitosis and meiosis. To understand the probable cause of aberrations, a simple study was performed during induced lymphocyte culture. The lymphocyte cultures were setup using the blood samples of cattle and buffaloes. Samples were harvested without adding ethedium bromide and colchicine. The Giemsa stained chromosome slides were screened under the microscope (100X) and photographed with attached

Merotelic kinetochore orientation occurs frequently during early mitosis in mammalian tissue cells and error correction is achieved by two different mechanisms.

Merotelic kinetochore orientation is an error that occurs when a single kinetochore becomes attached to microtubules from two spindle poles rather than just to one pole. We obtained the first evidence that merotelic kinetochore orientation occurs very frequently during early mitosis in mammalian tissue cells and that two different correction mechanisms are critical for accurate chromosome segre...

Merotelic kinetochore orientation versus chromosome mono-orientation in the origin of lagging chromosomes in human primary cells.

Defects in chromosome segregation play a critical role in producing genomic instability and aneuploidy, which are associated with congenital diseases and carcinogenesis. We recently provided evidence from immunofluorescence and electron microscopy studies that merotelic kinetochore orientation is a major mechanism for lagging chromosomes during mitosis in PtK1 cells. Here we investigate whether...

Screening of Breeding Bulls of Different Breeds through Karyotyping

A study of chromosomal analysis for 200 breeding bulls of different breeds of cattle (Jersey, Holstein Friesian, Sahiwal and Cross-bred) and Nili-Ravi buffalo, maintained at Semen Production Unit, Qadirabad and Livestock Experiment Station, Bhunikey (Pattoki) was carried out. Micromethod was adopted for leukocyte culture and chromosomes were trapped at metaphase stage. The diploid number of chr...

Cantharidin-induced mitotic arrest is associated with the formation of aberrant mitotic spindles and lagging chromosomes resulting, in part, from the suppression of PP2Aalpha.

Cantharidin, a natural vesicant, inhibits the activity of several PPP family phosphatases, displays antitumor activity, and induces apoptosis in many types of tumor cells. However, the molecular mechanisms underlying the antitumor activity of cantharidin are not clear. Here, dose-response studies confirm a strong correlation between the suppression of phosphatase activity and cell death. Flow c...
